Cryo-Ultramicrotomy enables to make thin sections of constant thickness (~20-200 nm) with very soft materials that cannot be cut at ambient temperature.
A cryo- chamber filled with liquid nitrogen is added to the ultramicrotome. Sample, knife and surrounding area are cooled down to the required temperature.
This method allows any sample orientation.
